How important is surface roughness (Ra) for GMP-compliant powder mixers, and which standards are achievable?

Surface roughness is a central GMP parameter for powder mixers. It directly influences cleanability, contamination risk, product behaviour and the qualifiability of the plant. For product-contact areas in pharmaceutical solids technology, Ra ≤ 0.8 µm typically applies as a proven standard. Depending on the process requirement, lower values are also used.

Significance of surface roughness in a GMP environment

A low and defined surface roughness is important for technical and regulatory reasons. Smooth surfaces can be cleaned and validated better. They reduce residues of powder, moisture and cleaning agents. This also reduces the risk of cross-contamination during product changes.

Smooth surfaces also improve hygienic safety. Rougher surfaces offer more adhesion points for particles and microorganisms. A good surface finish therefore supports a GMP-compliant design. It also helps in meeting the requirement for product contact surfaces that are easy to clean and non-absorbent.

Product quality benefits too. Cohesive powders adhere less strongly to smooth surfaces. This improves the yield, shortens discharge and supports reproducible batches.

Customary Ra values for GMP-compliant powder mixers

For standard applications in solids technology, Ra ≤ 0.8 µm is a customary guide value for product-contact surfaces. This value is usually achieved by mechanical grinding and subsequent fine polishing.

For higher demands on cleanability and product purity, Ra ≤ 0.6 µm or Ra ≤ 0.4 µm are often specified. Electropolished surfaces can be made smoother still. In particularly demanding applications, values of Ra ≤ 0.2 µm are also achievable.

Non-product-contact external surfaces usually lie at Ra 1.2 to 1.6 µm. There the emphasis is above all on a clean appearance and good wipe cleaning.

Standards and measurement

Roughness is generally stated as the arithmetic mean roughness value Ra in accordance with DIN EN ISO 4287/4288. For GMP documentation, measurement records produced with calibrated instruments or optical measuring systems are used.

Important guidance is provided by the EU GMP Guide, FDA requirements and EHEDG recommendations. ASME BPE is also frequently drawn upon as a reference in practice, even though it originally comes from liquid and bioprocess technology.

How amixon® solves surface finish and material selection for hygienic powder processes

Surfaces: ground, low in adhesion, cleanable

At amixon®, the mixing chamber and mixing tool are welded free of crevices and ground smooth – without scratches, pores or undercuts in which product residues could adhere or biofilms could form. The surface finish is established according to the application: from industrially ground to highly polished designs for GMP-critical processes, for instance in the pharmaceutical or baby food industry. Since every amixon® apparatus is a one-off in accordance with the URS, the required Ra value is defined and documented project-specifically. The surface roughness can be selected project-specifically, typically with Ra values of 0.8 µm, 0.6 µm, 0.4 µm or 0.2 µm; beyond this, amixon® can lap to a mirror finish and electropolish.

Smooth, homogeneous surfaces reduce build-up directly. Together with the microfine addition of liquids, the mixing chamber remains largely free of deposits even during moistening processes.

Range of materials

As a welding company with European, Japanese, Korean and American qualifications, amixon® processes, depending on the application, mild steel S355J2+N, Hardox as a wear-resistant material as well as austenitic stainless steels such as 1.4301, 1.4541, 1.4571, 1.4404, 1.4539 and 1.4529. The range is complemented by duplex stainless steels such as 1.4462, 1.4162 and 1.4363 as well as Alloy 59, Hastelloy-C22 and nickel for highly corrosive media. One reference is the vacuum mixer-dryer VMT 200 in a full design made of Alloy 59.

For abrasive products, hardened tools and ceramic plasma coatings are available as wear protection.

Constructional complement

The surface finish develops its full effect only in combination with freedom from dead space. The mixing tool is mounted at the top only and the discharge fittings have few dead spaces. Clever-Cut® inspection doors with an OmgaSeal® seal additionally support a hygienically compliant construction. The residual discharge of 99.997 % and better with free-flowing products reduces the quantity of residue that has to be cleaned. On request, the mixers meet the EHEDG and FDA hygiene guidelines and the 3-A Sanitary Standards.

Manufacture in Paderborn as the quality foundation

amixon® develops and manufactures exclusively at the Paderborn works – with a high depth of manufacture and components from Germany. As a certified welding company with European, Japanese, Korean and American qualifications, amixon® designs every apparatus as a one-off on the basis of the operator's URS. Quality control remains entirely in-house without gaps and every specification is traceable down to component level. This manufacturing autonomy also secures long-term supply, since components can be reproduced even after many years.

Qualifiable and documented

For regulated environments, amixon® supplies qualifiable plants. Every apparatus is a one-off manufactured in accordance with the URS in its own production, with quality control that has no gaps. amixon® assists on request with DQ, IQ and OQ. Documentation and execution follow, on request, EU GMP and FDA 21 CFR Part 11.

On request the mixers meet relevant standards such as EHEDG, the FDA hygiene guidelines, the 3-A Sanitary Standards, USDA, GMP, ATEX and ASME. They are integrated into the operator's validation concept from the URS through to commissioning.
